Well, the answer is no, on Instagram 2022, the app does not notify you when someone takes a screenshot in your direct message (DM).

So, they won’t be notified if you take a screenshot of your chat, in contrast.
You can take screenshots of your dm with someone and Instagram will never let him know that you did so.
However, the scenario is different if vanish mode is on
